Malaria parasites evolving in vaccinated laboratory mice become more virulent, according to research at Penn State University. The mice were injected with a critical component of several candidate human malaria vaccines that now are being evaluated in clinical trials. "Our research shows immunization with this particular type of malaria vaccine can create ecological conditions that favor the evolution of parasites that cause more severe disease in unvaccinated mice," said Andrew Read, Alumni Professor of Biological Sciences at Penn State.

"We are a long way from being able to assess the likelihood of this process occurring in humans, but our research suggests the need for vigilance. It is possible that more-virulent strains of malaria might evolve if a malaria vaccine goes into widespread use," Read said. The research, which will be published in the 31 July 2012 issue of the scientific journal PLoS Biology, showed that more-virulent malaria parasites evolved in response to vaccination, but the exact mechanism is still a mystery. It was not due to changes in the part of the parasite targeted by the vaccine.

No malaria vaccine ever has been approved for widespread use. "Effective malaria vaccines are notoriously difficult to develop because the malaria parasite is very complex. Hundreds of different malaria strains exist simultaneously within any local region where the disease is prevalent," Read said. Most vaccine developers use only small sections of the malaria parasite to produce an antigen molecule that then becomes a key ingredient in a highly purified malaria vaccine. Read's lab tested the antigen AMA-1, a component of several such vaccines now in various stages of clinical trials.

"Our laboratory experiments followed clues from theoretical studies and earlier experiments that suggested that some malaria vaccines could favor the evolution of more-virulent malaria parasites," Read said. If candidate vaccines do not completely eliminate all the malaria parasites, the parasites that remain have opportunities to evolve. A mosquito then could transfer the evolved parasite from the vaccinated person into a new host -- a process called leaking. "Leaky vaccines create a situation that further fosters parasite evolution," Read said.

The Penn State study found that parasites causing worse malaria symptoms in unvaccinated mice evolved after "leaking" consecutively through as few as 10 vaccinated mice. "The parasites that are able to survive in the immunized hosts must be stronger after having survived exposure to the vaccine," Read said. "The vaccine-induced immunity apparently removed the less virulent malaria parasites, but left the more virulent ones."

The AMA-1 antigen used in the Penn State study triggers the body to make anti-malaria antibodies. These antibodies recognize the AMA-1 antigen on the parasites and disable the malaria infection. The shape of the antigen ensures that the antibodies can bind securely with the malaria parasite -- like pieces in a jigsaw puzzle -- an important step in producing immunity. Scientists already knew that vaccines become obsolete when evolutionary mutations change the parasite's antigen structure in such a way that the antibody is not able to lock onto the targeted part of the parasite. But the Penn State study showed the malaria parasite evolved within the vaccinated mice even without any detectable changes in the antibody target on the parasite.

"We were surprised to find that more-virulent strains of malaria evolved even while the gene encoding the key antigen remained unchanged," said Victoria Barclay, the postdoctoral scholar in Read's lab who conducted the laboratory experiments and who is the corresponding author of the PLoS Biology paper. "We did not detect any changes in the gene sequence." The researchers conclude that evolution must have taken place somewhere in the parasite's genome. Read's lab now is hunting for the exact locations on the parasite's DNA where the mutations occurred.

"Generalizing from animal models is notoriously difficult in malaria," Read said, so the scientists do not yet know if this newly recognized type of evolution could happen in human malaria or with other rapidly evolving diseases, such as the viruses that cause AIDS or cervical cancer. "What we do know is that in Victoria Barclay's experiments in our lab at Penn State; with our parasites, our mice, and with this particular antigen; the malaria parasites that evolved through vaccinated hosts become more virulent," he said.

"Vaccines are one of the most fantastically cost-effective health gains we've ever had, so there is no question that we should proceed on all fronts to develop a safe and effective vaccine against malaria," Read said. "At the same time, our research is revealing new reasons to proceed with vigilant caution." Read suggests that vaccine researchers conducting clinical trials should not only be carefully monitoring for parasite evolution at the vaccine target, but they also should watch for mutations throughout the parasite's entire genome. "This sort of monitoring also should go on once a new vaccine goes into widespread use," he said. "It appears that in a world with leaky vaccines, virulent pathogen strains can evolve. Different vaccines or other transmission-blocking measures might be needed to stop the spread of any evolved parasites," Read said.

Insurance premiums may increase to ensure flood-hit homeowners get

UK householders face higher home insurance premiums so that those in high flood risk areas can still get cover.

The Association of British Insurers (ABI) says that the insurance industry and the government are discussing a scheme to ensure that 200,000 households affected by flooding will be able to renew their policies next year.

The scheme, proposed by the ABI, means a cap on premiums. Any house that would normally incur a much higher premium because of flood risk will have the extra paid out of a levy on every home policy in the UK.

Insurers agreed a statement of principles with the government in 2008 to renew cover ? usually at a very high premium and with an even higher excess ? for flood victims. But this agreement runs out on 30 June 2013, and householders then face being refused insurance unless the government and the industry come to a new agreement.

A spokesman for one insurer said that providing cover for people in high-risk areas such as Hebden Bridge, which has flooded three times in as many weeks, was becoming assurance rather than insurance; covering an event that would definitely happen, rather than a risk.

He added: There are policies being written now that will not be offered renewal next year unless the government acts soon.

The ABI said a scheme called Project Noah, which would involve reinsuring flood risk on the international market, had now been discounted.

A government spokesman said: We have not yet arrived at an agreement, but discussions with the insurance industry continue to be constructive and we are working together to reach a solution that is practical and affordable for households. Were mindful of those on lower incomes living with the risk of flooding. Thats why were considering ways to keep flood insurance affordable for those who might struggle most with premium increases.

The ABI estimates that damage caused by the floods in the last few weeks amounts to between ?200m and ?300m.

(AP) ? Authorities say a Kent State University student is due in court after allegedly posting a message on Twitter saying he planned to "shoot up" the northeastern Ohio campus.

Nineteen-year-old William Koberna was arrested at his parents' home in the Cleveland suburb of Brunswick Sunday afternoon. The sophomore has been charged with inducing panic, a felony, and aggravated menacing. He was to be arraigned Monday.

A university spokesman says the tweet was posted July 25.

University officials monitoring Twitter last week noticed the profanity-laced message and contacted police. They said it included a threat that "I'm shooting up your school ASAP."

Kent State University President Lester Lefton said in a statement that "our students, employees, and all those who come to campus should know that their safety is our top priority."

A senior Haryana IAS officer has hinted at an alleged nexus between politicians and bureaucrats in a case of transferring prime land in Gurgaon ? belonging to gram panchayats and the government ? to individuals and real-estate firms.

Director General of Consolidation Holdings (Haryana) Ashok Khemka, in a letter to the Additional Chief Secretary and Financial Commissioner (Revenue), has recommended the registration of a criminal case against two senior Haryana IAS officers ? S C Goel and Anil Kumar. He also suggested a thorough probe be conducted by the Central Bureau of Investigation (CBI).

In the letter (copy with The Indian Express) dated July 25, Khemka highlights 20 cases in which real-estate firms and private individuals were given prime land in Gurgaon (mainly Sectors 58-63 and Sector 65-67), through orders issued by Anil Kumar and SC Goel. The cases also reveal that consolidation proceedings of the land under question were completed 40 to 45 years ago, but had only now been transferred.

Khemka?s letter reads that the beneficiaries in whose favour land was transferred, ?would not have even been in existence, during the pendency of the consolidation proceedings?.

Calling it a ?gross abuse of position by the authority concerned?, Khemka hinted at the possibility of several other such orders that were passed by other officers.

House Ways and Means chairman fighting cancer

WASHINGTON (AP) ? The chairman of the House's tax-writing committee says he's been diagnosed with a "very early, highly treatable and curable type" of cancer.

GOP Rep. Dave Camp of Michigan says in a statement that doctors found non-Hodgkin lymphoma during a recent physical. Camp says he'll continue in Congress and retain his committee chairmanship during chemotherapy.

Non-Hodgkin lymphoma is a cancer that begins in the cells of the immune system. The 59-year-old Camp says he has large B-cell lymphoma. B-cells are a type of white blood cell that helps fight infections.

Camp, who's served since 1991, has said he wants to combine a push to head off pending tax increases with ways to force Congress to revamp the tax code next year.

Summer Wedding Wreath with Ribbon Roses Tutorial

When I started this today, the sun was shining and I thought any Bride would be happy with the day... but then the rains came, then the thunder and lightening, and finally, to round it off we had hail. ?The UK will certainly take the weather gold medal this weekend, if only for diversity!
One of the things I always remember about wedding is not so much the dress but the little touches, which is why this wreath is made from fabrics you might be making your bride and bridesmaid's dresses from. ?I hit my Jane Means stash of ribbons to start off this week's tutorial, so get yourself ready and let's get started:


You will need:
A polystyrene form
Half a yard / metre each of ?net, organza or sari fabric (basically a sheer weave fine fabric)
Rotary cutter with pinking wheel and cutting mat or pinking shears
Chopstick
2/3 yards or metres of organza wired ribbon?
Household scissors
Matches
Tweezers or small pliers
2 metres of plain ribbon
Well we have made wreathes at Christmas using this technique last year but this is a little bit different as the wreath is a little more blousey.
Start by cutting out three strips, three inches wide, then cut across them at three inches to form three inch squares. I found some fabulous organza with a sparkle diamante type detail.
Using your chop stick in the centre of the square, push it into the polystyrene form.
Continue to build it up, cutting more squares as you need them, you don't need to cover every inch... walk away from the wreath and turn it round as you can become a touch anal about there being no white showing!
When you are happy that it is covered, go an make yourself a drink and take a final look.
Something like this will do :)
Now cut your ribbon, you will need lengths of about 18", use you household / kitchen scissors as you will be cutting through wire and I will personally come to your house and remove all your dressmaking scissors if I catch you blunting them on wired ribbons! ?Remember to flame the ends with your matches to seal them.

You will find fine wire running through the top and the bottom of your ribbon, gently tease the ends out of the top of the ribbon and pull carefully to start gathering the ribbon... do not tug, the wire is fine and will break...
Now from the ungathered side fold over one end, which will tuck it in when you start to roll up your rose. ?Now you could use a match to wind the ribbon around but I find it easier to wind it free hand.
?You can manipulate your flower when you have finished winding.
Twist the two wires together a few times to secure your rose, then separate them.
Place the rose on your wreath, manipulate your wire through the organza and net, to the back of the wreath and use your pliers to twist the wire to hold the rose in place. ?Tidy in the ends and cut away the excess. ?Repeat the process to add as many roses as you wish.
Now use a loop of ribbon to create your bow at the top of the wreath and pop it on the front door to show everyone where the festivities are starting from.
And there you have it, a souvenir of a very special day.

Anaheim Police Shooting: Officers Fire At Suspected Burglars After

ANAHEIM, Calif. -- Anaheim police opened fire on a burglary suspect Friday morning, days after officer-involved shootings killed two people and sparked violent protests in the Southern California city.

It was unclear whether anyone was hurt in the confrontation.

"We have been in contact with the local hospitals. There is no indication that anyone has come in with a gunshot wound," Sgt. Bob Dunn said.

Police answering a burglar alarm at a condominium complex clubhouse at about 3:15 a.m. spotted appliances outside and a man leaving the building, Dunn said.

An officer began chasing him while another officer checked out a nearby car, which suddenly sped off in the direction of the first officer, Dunn said.

The second officer then opened fire on the car but the driver escaped, Dunn said.

A search for the first man ended about 45 minutes later when a paroled burglar was found hiding near some train tracks, Dunn said.

The man had been bitten by a police dog and was taken to a hospital for treatment before being booked on suspicion of burglary and resisting arrest. His name was not immediately released.

The shooting came as activists in Los Angeles, Seattle and other cities called for Friday solidarity protests over the earlier police killings.

Last Saturday, a police officer fatally shot Manuel Diaz outside an apartment complex. Officers said Diaz, who had a criminal record, failed to heed orders and threw something as he fled police.

The city's police union said Diaz reached for his waistband, which led the officer to believe he was drawing a gun.

Diaz's family, which is suing for $50 million in damages, says he was shot in the leg and the back of the head.

On Sunday night, police shot to death Joel Acevedo, a suspected gang member they say fired at officers after a pursuit.

The shootings prompted four days of violent protests, culminating Tuesday night in hundreds of demonstrators surging through downtown streets. ST. LOUIS - Hanley Ramirez gave the Los Angeles Dodgers a good look at what he can bring to the table Wednesday night.

Ramirez, traded from Miami earlier in the day, went 2 for 4 with a walk, an RBI and a run scored in his debut, but it wasn't enough to lift the Dodgers in a 3-2, 12-inning loss to the St. Louis Cardinals.

Still, Ramirez felt good about his first game with a new team.

"I feel comfortable," Ramirez said. "Great group of guys here; you just go out there and have fun. I'm happy to be here."

Hoping for a smooth transition, Ramirez started at third base. After moving from shortstop when the Marlins signed Jose Reyes, Dodgers manager Don Mattingly said Ramirez will return to his old position as soon as he's comfortable with a switch.

Ramirez took to wearing the Dodger blue right away, hitting a triple off the center-field wall in his first at-bat.

"I thought it was a pop-up to center to be honest," Ramirez said. "It just kept going and going."

Los Angeles acquired the three-time All-Star and former NL batting champion along with reliever Randy Choate, while the Marlins received pitcher Nathan Eovaldi and minor league pitcher Scott McGough.

Choate pitched a scoreless inning in relief in his debut.

"The extra innings get you in the game," Choate said. "It's just been kind of a long day."

Mattingly's day ended a little early when he was ejected in the 10th inning by home plate umpire Jeff Kellogg for arguing balls and strikes. Mattingly was still upset afterward.

"Nothing really happened," he said. "I was just getting frustrated. The strike zone was horrible."

The 28-year-old Ramirez said before the game he didn't care where he played, emphasizing how happy he is to join a contender. He declined to blame his off-year offensively on the position switch and agreed with suggestions that this was a fresh start.

He was batting .246 with 14 homers and 48 RBIs in 93 games coming in.

"Yeah, why not? It's a new beginning," Ramirez said. "I've just got to go out there and have fun."

Ramirez's new teammates sensed the commitment from ownership to keep charging. The Dodgers have won five of seven and are 2 1/2 games behind NL West-leading San Francisco.

"Yeah, it's no excuses now," lefty ace Clayton Kershaw said. "Last year we had excuse, excuse, excuse and we ended up playing pretty well in the second half, and this year it's pretty much all on us."

Mattingly has been using a combination of Juan Uribe, Adam Kennedy and Jerry Hairston Jr. at third base, and career minor leaguer Luis Cruz has been starting at shortstop in place of injured Dee Gordon.

"It works best for us right now at short," Mattingly said. "The best answer I got from Hanley was, `It doesn't matter. Third or short, I just want to win."'

Mattingly added that he'd be patient with Ramirez.

"It's really just up to him as far as I'm concerned, it's not something I'm going to see that he's ready," the manager said. "It's going to be more like he tells me he's ready, I'm playing short."

After getting announced to zero response from a near-sellout crowd for his first at-bat in the second, Ramirez tripled against Kyle Lohse and scored on a sacrifice fly by James Loney to give the Dodgers an early lead.

Ramirez's bat gives Mattingly a big three to anchor the lineup along with Matt Kemp and Andre Ethier. Kemp found out about the trade sitting in bed checking Twitter.

"I was like `Whoa, that can't be true,"' Kemp said. "And then I saw more tweets and more and I was like `Wow, I guess this is true."'

Prior to the trade, the Dodgers were in the bottom half of the National League in most offensive categories.

Mattingly advised Ramirez to take a deep breath and just be himself. He said he wouldn't worry about whatever happened in the past

"I'm sure he's going to want to show people he can play," Mattingly said. "But we'll be mindful of just reminding him that we just want him to play.

"Have good at-bats, play the game, and do his thing. That's all we'll ask."

The first reason I believe why people do not set goals is they do not see its importance. Like people who do not develop their communication and other aspects of themselves, they do not see the importance of setting goals. I have met people who are unbelievably resistant to developing their communication. They have actually found it insulting to think they need to improve how they communicate! Every single person on the face of this Earth can always communicate more effectively and have their life improve as a result. Likewise, with goals there are people who do not see the importance of setting personal goals. Goal-setting forms the foundations for personal achievement and it is of the utmost importance.

The second reason why people do not set goals is a fear of others caused from criticism. People criticize because they see faults in others or are brainwashed by society and other beliefs into thinking greatness cannot be achieved. They criticize to hold people back from achieving goals. Average people do not want others to go beyond average. A person can fear setting goals because each time they have set goals in the past, others have criticized and shot down their dreams. Others impose their own self-limiting beliefs through criticism instilling fear within the person. To bluntly solve this, screw what people think. Screw allowing people to destroy your life with their beliefs. Society is filled with endless garbage to prevent you from achieving your goals. If you want something, go and get it by setting and achieving personal goals.

The third reason why people do not set goals is a fear of failure. They can think success is derived from not failing. The opposite is true. Success comes from failing a lot and failing fast. Famous inventor Thomas Edison said, ?I haven?t failed. I?ve just found 10,000 ways that won?t work.? Being an inventor, Edison understood that failure was apart of success. Failure was the nuts and bolts that constructed his success. Understand that failure occurs and it shows you are taking action. Learn from failure and enjoy knowing you are moving towards your goals instead of fearing to set out after them in the first place.

The fourth reason why people do not set goals is a fear success. People who fear success believe it is bad. The fear is common in wealth and wanting an attractive partner because negative results will occur. They believe wealth is evil or a certain type of partner is unfaithful. The fear prevents them achieving what they want by not laying the foundations of their desire with goal-setting.

The fifth reason why people do not set goals is they do not know how. This reason relates to fear of failure where the person has set goals in the past, but has not achieved them and becomes discouraged from the failure. These people become disheartened from the failure and think failure is imminent. They do not learn from past mistakes and move towards personal growth. There are techniques such as setting SMART achievable goals which I have written about but are not covered in this article. Using SMART goals enables you to set and achieve goals then merely wishing a result based on temporary motivation too common around the new years period.

The sixth reason why people do not set goals is they lack the inner desire to want something greater than what they currently have. For them, the ordinary or even sub-ordinary is sufficient. These people do not believe they can reach or deserve success because society has conditioned them into thinking they are ordinary people and therefore must do ordinary things. It can also be caused by failure, criticism, and other reasons as to why people do not set goals.

Swiss Study: Colonoscopy with Polypectomy Reduces Cancer

OAK BROOK, Ill.?A study from researchers in Switzerland found that colonoscopy with polypectomy significantly reduces colorectal cancer incidence and colorectal cancer-related death in the general population.

A total of 12 colorectal cancer cases were identified in the screening group of 1,912 patients and 213 cases of colorectal cancer were found in the non-screened group of 20,774 patients. One of the 12 persons of the screened individuals with a colorectal cancer and 51 of the 213 persons of the non-screened individuals with a colorectal cancer died because of their cancers. The study appears in the July issue of GIE: Gastrointestinal Endoscopy, the monthly peer-reviewed scientific journal of the American Society for Gastrointestinal Endoscopy (ASGE).

Colorectal cancer (CRC) has a very high incidence in Switzerland as well as in other European countries and is the second most frequent cause of cancer-related deaths in Europe. It is detected in approximately 413,000 people in Europe every year, half of whom die because of the disease. Therefore a need exists for efficient strategies for prevention and early detection of CRC. Colonoscopy with the possibility of an immediate polypectomy is a recommended and preferred screening method because polyps (growths in the colon) can turn into cancer over the course of years to decades. Removing polyps during a colonoscopy prevents that polyp from becoming cancerous.

?In contrast to earlier CRC screening studies that used colonoscopy, this population-based closed cohort observational study aimed to obtain complete and comparable data on CRC incidence and CRC-related mortality after a single screening colonoscopy compared with no screening, while taking into account the potential differences in risk profiles between the screened and non-screened participants," said study lead author Urs A. HTC to build another Facebook phone in 2013, says Bloomberg

Despite failing to set the world alight with 2011's ChaCha and Salsa, HTC and Facebook will collaborate once again to bring another Facebook phone to market next year, according to reports from Bloomberg. ?Details on the purported device are scant at the moment, but the report, which cites the usual "people with knowledge of the matter," indicates a mid-2013 release, after the launch was apparently pushed back from the end of 2012.

The site doesn't mention Android specifically, but we'd be surprised if Google's OS wasn't involved in some way or other, given the history of collaborations between Facebook and HTC. Bloomberg? says the phone will run a "modified" operating system, which suggests to us that we may see a forked version of Android, similar to what Amazon's done with the Kindle Fire. For the less technically inclined, that means a version of the OS built upon the open-source code, and most likely excluding any Google apps.

Android followers will remember that last year's HTC ChaCha (aka AT&T HTC Status) and its Europe-only sibling, the Salsa, weren't met with much success. So it's possible we'll see the two companies adopt a different strategy this time around. With Facebook getting more involved in the app space, it'd make sense for the Facebook phone to be heavily invested in this, rather than something like Google Play.

In any case, it seems the rumored device is still the best part of a year away from completion, so it's possible we'll see more changes throughout the course of development.
